PUR-1 is essentially non-fluorescent in the absence of nucleic acids. Its fluorescence is significantly enhanced in the presence of DNA and/or RNA. In the presence of RNA, PUR-1 has fluorescence at ~478 nm with fluorescence quantum yield of ~40%. PUR-1 dye is excitable at 488 nm (with maximal excitation at 460 nm) and emits fluorescence between 470 to 550 nm with a maximum emission at 478 nm in the presence of nucleic acids. The dye can be used for analyzing blood-borne parasite infections in combination with a flow cytometer.
