Graph illustrates signal-to-noise ratio (SNR) x 100%. ATP dose response was measured in CHO-K1 cells with Screen Quest™ Calbryte 590 Probenecid-Free and Wash-Free Calcium Assay Kit. CHO-K1 cells were seeded overnight at 50,000 cells/100 µL/well in a 96-well black wall/clear bottom costar plate. 100 µL dye loading solution was added and incubated for 45 min at 37°C and 15 min at RT. ATP (50 µL/well) was added by FlexStation 3 to achieve the final indicated concentrations.

Overview

Screen Quest™ Calbryte™ 590 Probenecid-Free and Wash-Free Calcium Assay Kit is designed for red fluorescence-based detection of intracellular calcium mobilization in live cells. Calcium flux assays are widely used in drug discovery for screening G protein-coupled receptors (GPCRs) and calcium channels. The kit uses Calbryte™ 590 AM, a cell-permeant calcium indicator that enters cells and is converted by intracellular esterases into a calcium-responsive fluorescent dye.

Calbryte™ 590 AM is designed for improved cellular retention, enabling calcium flux detection without probenecid and without wash steps. Upon cell stimulation, receptor-mediated calcium release or calcium influx produces an increase in red fluorescence that can be measured in a microplate format. Compared with green fluorescence-based calcium assays, the red fluorescence readout can help reduce interference from colored compounds in screening libraries and is compatible with GFP-expressing cell lines for high-content analysis. Screen Quest™ Calbryte™ 590 Probenecid-Free and Wash-Free Calcium Assay Kit is suitable for GPCR calcium signaling studies, calcium channel assays, GFP-compatible calcium imaging, high-throughput screening, and calcium flux analysis in fragile or difficult cell lines.
