What is the difference between methanol and formaldehyde fixation?

Answer

Alcohol based fixations dehydrate cells/tissues, causing proteins to denature and precipitate in situ, however small molecules may be lost during the subsequent steps of fixation. Paraformaldehyde causes covalent cross-links between molecules, effectively gluing them together into an insoluble meshwork.
