Cyclic Nucleotide PDE Substrates

These fluorescent cyclic nucleotide derivatives serve as specific substrates for PDE IV (cAMP-specific) and PDE V (cGMP-specific) enzymes. Upon hydrolysis by the target phosphodiesterase, the substrates release fluorescent products that can be quantified using standard fluorescence plate readers. Both green (FAM) and red (TAMRA) fluorescent options are available to accommodate different instrument configurations and multiplexing needs.

Glycerophosphocholine PDE Substrate

Resorufin phosphocholine is a fluorogenic substrate for glycerophosphocholine phosphodiesterase. The enzyme cleaves phosphocholine from the substrate, releasing highly fluorescent resorufin (Ex/Em = 571/584 nm) that can be measured with standard red fluorescence filter sets. This substrate provides a convenient, real-time assay for monitoring phosphocholine release activity.
Calibrators and Standards

Fluorescein-conjugated nucleotide monophosphate calibrators enable accurate quantification of PDE activity by providing reference standards that mimic the hydrolysis products. Use AMP-Fluorescein with FAM-cAMP substrates and GMP-Fluorescein with FAM-cGMP substrates to generate standard curves for converting fluorescence signals to enzyme activity units.
Technical Considerations

When selecting a PDE assay system, consider the following factors:
Substrate Specificity
  • FAM-cAMP substrates are suitable for cAMP-specific PDEs (PDE4, PDE7, PDE8) and dual-specificity PDEs (PDE1, PDE2, PDE3, PDE10, PDE11)
  • FAM-cGMP substrates are appropriate for cGMP-specific PDEs (PDE5, PDE6, PDE9) and dual-specificity enzymes
  • Choose TAMRA-labeled substrates when sample autofluorescence in the green channel is problematic
Assay Optimization Tips
  • Titrate enzyme concentration to achieve 50-70% substrate conversion for optimal Z' values
  • Include appropriate controls: no enzyme, no substrate, and known inhibitor standards
  • Consider compound interference by running counter-screens with fluorescent standards
