Current demands for RT-PCR testing of SARS-CoV-2 have led to a worldwide shortage of RNA extraction kits. A key missing component required by extraction kit manufacturers and hospitals is the RNA Lysis buffer. This ReadiUse™ Viral RNA Lysis Buffer can be used as an alternative to the Roche MagNA Pure 96 External Lysis Buffer (Code: 06374913001), Biomérieux NucliSENS Lysis Buffer (Code: 280134), and Qiagen Buffer RNAeasy Lysis Buffer (Code: 79216).
Example protocol
PREPARATION OF WORKING SOLUTION
ReadiUse™ Viral RNA Lysis Buffer working solution
Add 0.5 mL of β-Mercaptoethanol to 50 mL of ReadiUse™ Viral RNA Lysis Buffer
Note: Store the RNA Lysis Buffer containing β-Mercaptoethanol at 4 °C.
SAMPLE EXPERIMENTAL PROTOCOL
The following protocol can be used as a guideline and should be optimized according to your experimental needs. This protocol was optimized for use with at least 1.5 X 103 cells to a maximum of 5 X 106 cells per purification.
- Wash the cells once with sterile 1X PBS.
- Add 100 µL of ReadiUse™ Viral RNA Lysis Buffer working solution to the washed cells. Mix by pipetting.
- Purify extracted RNA from the previous step using RNA purification method of choice.
