ReadiUse™ SATA (N-succinimidyl S-acetylthioacetate) is an NHS ester-based thiolation reagent used for introducing protected sulfhydryl groups onto proteins and other primary amine-containing biomolecules. The NHS ester reacts with lysine side chains and N-terminal amino groups to form stable covalent amide bonds while introducing an acetyl-protected sulfhydryl functionality. The protected thiol group can subsequently be deprotected using hydroxylamine to generate a free sulfhydryl group for downstream conjugation reactions.

The protected sulfhydryl strategy enables preparation and storage of modified biomolecules prior to thiol exposure and conjugation. ReadiUse™ SATA can be used for thiolation of antibodies, proteins, peptides, and other amino-containing biomolecules in crosslinking, labeling, immobilization, and bioconjugation workflows. Following deprotection, the exposed sulfhydryl groups can participate in thiol-reactive chemistries including maleimide-based conjugation and disulfide bond formation.
