xtraFluor™ Violet 420 Amine
xtraFluor™ Violet 420 Amine is a violet laser-excitable, bright fluorophore designed for flexible thiol-reactive conjugation workflows using SMCC based in situ maleimide generation.
  • Violet laser optimized: Ex/Em ≈ 407/424 nm with efficient excitation using standard 405 nm flow cytometry lasers
  • In situ maleimide generation: Primary amine functionality enables formation of thiol-reactive maleimide-functional XV420 intermediates using SMCC crosslinking chemistry
  • Bright violet emission: Narrow emission profile supports reduced spillover and efficient spectral unmixing in multicolor panels
  • Flexible conjugation workflow: Suitable for thiol-reactive labeling of reduced antibodies, peptides, and proteins
  • Improved dim population resolution: Higher brightness than Pacific Blue and BD Horizon V450 with spectral compatibility to BV421
(Top) Spectral emission profiles generated using four spatially offset lasers (355 nm, 405 nm, 488 nm, and 640 nm). Each laser produced a distinct emission pattern, and their combination yielded the composite spectral signature.
(Bottom) Flow cytometry analysis of whole blood stained with xtraFluor™ Violet 420-labeled Anti-human CD4 Antibody (Clone: RPA-T4) (Cat. #10041000) prepared using xtraFluor™ Violet 420 Amine (Cat. #70806). The fluorescence signal was monitored using Cytek Aurora spectral flow cytometer in the V1-A channel. The antibody was used at 0.5 µg/test.
