Spectrophotometry is a method used to estimate the concentration of analyte in a solution by measuring intensity of light as a beam of light passes through the solution. It works on the principle that every material absorbs light of a specific wavelength as it passes through the solution.

The relationship between concentration and absorbance is best explained by the Beer-Lambert law, which relates the attenuation of light to the properties of a material. The Beer-Lambert law states that the concentration of a chemical solution is directly proportional to its absorption of light. There is a linear relationship between the concentration and the absorbance of the solution, which enables the concentration of a solution to be calculated by measuring its absorbance.
