Affinity chromatography is the most selective and robust chromatography technique and is considered to be the best for protein purification. This technique gives the purest results and is often used to complete the protein purification process.

Affinity chromatography works by manipulating the high affinity of many proteins for a specific chemical group. Under favorable conditions, different types of proteins exhibit highly specific interactions with particular ligands. Affinity chromatography uses this property of proteins to adsorb the target protein from the extract as it passes through a column packed with an appropriate adsorbent. The unwanted substances are washed away and the protein of interest is then eluted and made available to be used in a wide range of downstream applications.

Affinity purification is also called affinity chromatography.
