How do I prepare culture media?

The steps to prepare culture media are described below. 

  1. Mix and Dissolve: Add water (up to 80% of the required volume) to the powdered medium to rehydrate it. Stir well to dissolve the medium components completely. If preparing an agar medium, heat and stir until the agar powder is dissolved. 
  2. Prepare Tubes or Plates: Distribute the prepared liquid medium into tubes or containers for culturing microorganisms. If making agar plates, pour the agar medium into sterile petri dishes.
  3. Sterilization: Use an autoclave to sterilize the media in tubes. For an agar plate, autoclave the medium before pouring it into sterile petri dishes. 
  4. Cooling and Solidifying: Allow the sterilized agar medium to cool to suitable temperature, allowing it to solidify in the petri dishes.
