What are the advantages of native ChIP?

Answer

Native ChIP is suitable for exploring histone marks and abundant targets. An additional advantage is that no chemical fixation of cells is required, as cells remain in a more natural state. Additionally, when using native ChIP, there may be an increased affinity of antibody binding to antigens on native chromatin because it is more accessible, (antisera are typically raised against unfixed proteins). Another advantage is that the immunoprecipitation is highly efficient, and often yields sufficient material for protein analysis. Because of this, PCR amplification is not required to analyze the precipitated DNA. Additionally, antisera are typically raised against unfixed proteins, thus the specificity of the antibody binding to unfixed chromatin is more predictable.
