What are the limitations of Illumina sequencing?

Answer

There are various limitations of illumina. One main limitation is sample loading sensitivity. Overloading the sequencing platform with too much DNA can lead to issues such as overlapping clusters and reduced sequencing quality. Consequently, the overall error rate of this sequencing technology is approximately 1%. Another limitation is related to fragment size, primarily because of the sequencing procedure that involves attaching short DNA stretches to a glass plate. Larger DNA fragments exhibit difficulties in attachment, leading to less accurate sequencing. Lastly, one must wait until the sequencing process is finished before gaining access to the generated sequence data. Real-time data is not available.
