What are the steps for transfection with calcium phosphate?

Answer

Calcium phosphate transfection is a widely used technique for introducing foreign DNA plasmids into cells. Transfection with calcium phosphate follows these steps: 

  • DNA is mixed with the transfection agent – calcium chloride. 
  • The DNA and Calcium Chloride complex is added to phosphate buffer to generate nanoparticulate precipitates. Optimum pH range is between 6.9 and 7.4.
  • The DNA and Calcium Chloride complex precipitates. 
  • The precipitate is removed and dispersed in cell culture.
  • Phagocytosis of the co-precipitate by the target cell.
