Calcium phosphate transfection is a widely used technique for introducing foreign DNA plasmids into cells. Transfection with calcium phosphate follows these steps:
- DNA is mixed with the transfection agent – calcium chloride.
- The DNA and Calcium Chloride complex is added to phosphate buffer to generate nanoparticulate precipitates. Optimum pH range is between 6.9 and 7.4.
- The DNA and Calcium Chloride complex precipitates.
- The precipitate is removed and dispersed in cell culture.
- Phagocytosis of the co-precipitate by the target cell.
