What are the advantages of direct detection methods in immunohistochemistry (IHC)?

Answer
  • The process eliminates the need for a secondary reagent and an additional incubation step. 
  • It is a faster process overall because of the fewer steps involved.
  • The primary antibody can be labeled and conjugated directly to an appropriate fluorochrome or enzyme such as alkaline phosphatase or horseradish peroxidase
  • There is a smaller chance of a non-specific signal. 
  • Designing multicolor experiments is easy because a wide variety of fluorochromes is easily available. 
  • Is the recommended technique for detecting highly expressed antigens.
