What are the advantages of the labeled streptavidin-biotin (LSAB) method?

LSAB systems allow the primary antibody to be diluted, which can help optimize the staining conditions and minimize non-specific binding while still achieving robust signal detection. LSAB complexes also utilize streptavidin, which has lower non-specific binding compared to avidin. This reduces background staining. LSAB complexes are also more stable than ABC. Streptavidin-biotin interactions are highly stable, providing greater resistance to dissociation compared to avidin-biotin interactions. Additionally,  Streptavidin-biotin interactions allow for multiple biotin molecules to bind to each streptavidin molecule, leading to strong signal amplification.
