What are the disadvantages of direct detection methods in immunohistochemistry (IHC)?

Answer
  • The binding between the antibody and the target protein may be affected by the coupling of the label to the primary antibody
  • It is less sensitive than the indirect detection technique and may be better suited for applications in which the primary antibody is highly specific and there is a sufficient amount of target proteins. 
  • Every primary antibody needs to be labeled, which can add to the cost and time.  
  • It cannot be used for other applications down the line because the primary antibody is tagged with a label.
