About
Protein Kinases
Protein kinases catalyze the transfer of phosphate groups from ATP to specific substrates, playing critical roles in cell signaling, metabolism, and disease pathways. AAT Bioquest offers protein kinase activity assay kits and associated reagents that measure ATP-to-ADP conversion resulting from substrate phosphorylation, a characteristic outcome of protein kinase reactions, enabling antibody-free and non-radioactive detection of kinase activity under appropriate assay conditions.

Universal Kinase Activity Assays

Protein kinases transfer phosphate groups from ATP to specific protein substrates, generating ADP as a universal byproduct. Measuring ADP accumulation provides a direct, antibody-free method for detecting any kinase activity. AAT Bioquest offers two complementary assay platforms for universal kinase detection:
  • Amplite® Universal Fluorimetric Kinase Assay Kit — A complete, optimized kit specifically designed for protein kinase activity measurement and inhibitor screening
  • PhosphoWorks™ Fluorimetric ADP Assay Kit — A versatile ADP detection kit suitable for kinase assays as well as ATPase and other ADP-generating enzyme assays
Both kits convert ADP to a red fluorescent signal through enzyme-coupled reactions, enabling sensitive and quantitative measurement in microplate format. This approach offers several advantages over traditional methods:
  • Universal detection — Works with broad range of protein kinases regardless of substrate specificity
  • No antibodies required — Eliminates the need for phospho-specific antibodies
  • No radioactivity — Safer alternative to radiolabeled ATP assays
  • Homogeneous format — Simple add-and-read protocol with no wash steps

These supporting reagents enable researchers to optimize and control their kinase experiments. Staurosporine is a broad-spectrum kinase inhibitor commonly used as a positive control for kinase inhibition studies and as an apoptosis inducer. The ReadiUse™ Universal Kinase Reaction Stopping Buffer effectively terminates kinase reactions by chelating essential divalent metal ions (Mg²⁺, Mn²⁺), thereby preventing further ATP binding and phosphate transfer, providing consistent endpoint measurements across multiple samples.
Key Features
  • ReadiUse™ format — Pre-formulated solutions eliminate preparation time and improve reproducibility
  • Staurosporine stock solution — 1 mM concentration in DMSO, ready for immediate use in cell-based or biochemical assays
  • Stopping buffer — Metal ion chelation ensures complete and uniform reaction termination
