AAT Bioquest offers a comprehensive selection of protein quantification assays and reagents designed to meet diverse research needs. Our Amplite® assay kits provide ready-to-use solutions for measuring total protein concentration using established methods including BCA, Bradford, and fluorescent detection. In addition, we offer kits for protein characterization, including quantification of thiol, aldehyde and maleimide surface modifications.
Protein Functional Group Quantification (Thiols, Aldehydes)
Amplite® kits (colorimetric, fluorimetric)
Total Protein Quantification
AAT Bioquest's Amplite® protein quantification kits offer multiple detection methods to accommodate different sample types and instrumentations. The BCA (bicinchoninic acid) assay detects reduced copper ions generated by protein peptide bonds, while the Bradford assay uses Coomassie dye binding to aromatic amino acids. Amplite® fluorimetric assays using our proprietary orange-fluorescent dye, Prolite™ Orange, provide enhanced sensitivity for samples with limited material.
Key Features
No heating required — Amplite® BCA assay works at room temperature, unlike traditional BCA methods requiring 37 °C or 60 °C incubation
Broad linear range — Detection from sub-microgram to milligram protein quantities
Multiple protein compatibility — Validated with BSA, ovalbumin, thyroglobulin, and other common proteins
Flexible formats — Compatible with 96-well and 384-well microplates
Portelite™ compatibility — Fluorimetric kit optimized for CytoCite™ and Qubit™ fluorometers
Fluorescamine is a classic amine-reactive fluorogenic reagent widely used for protein and peptide quantitation. The compound is intrinsically non-fluorescent but reacts rapidly with primary aliphatic amines to produce highly fluorescent products. This reagent is ideal for researchers who prefer to develop custom assay protocols or need a versatile amine detection tool for various applications including HPLC detection and gel staining.
Protein Functional Group Quantification (Thiols, Aldehydes)
Site-selective protein modification is essential for bioconjugation, drug development, and biochemical research, and these assays are designed to quantify reactive functional groups rather than total protein concentration. AAT Bioquest provides specialized kits for quantifying specific functional groups on proteins, including aldehyde groups introduced through periodate oxidation and maleimide groups used in thiol-maleimide conjugation chemistry. These assays are particularly valuable for characterizing proteins for conjugation, validating bioconjugation reactions, and quality control of modified proteins.
Key Features
Aldehyde quantitation — Measures aldehyde content on oxidized glycoproteins for hydrazide/aminooxy conjugation
PE-Maleimide quantitation — Determines maleimide incorporation for phycoerythrin conjugate preparation
Simple colorimetric readout — No specialized equipment beyond standard microplate reader required
Rapid protocols — Results within minutes at room temperature
