DY677 CEP (DyLight 677 phosphoramidite) is an excellent replacement to the commonly used Cy5.5 phosphoramidite. Cyanine phosphoramidites (such as Cy3 and Cy5 phosphoramidites) are the most popular fluorescent labeling reagents for preparing fluorescence-labeled oligonucleotides. However, both Cy3 and Cy5 phosphoramidites are not very stable. DY677 CEP appears to be more stable than Cy5.5 phosphoramidite. As Cy5.5 phosphoramidite, the deprotection of oligos containing DY677 CEP may be carried out with ammonium hydroxide at room temperature, regardless of the base protecting groups on the monomers used. However, it is always prudent to use monomers with base labile protecting groups to limit the exposure time to 2 hours or less at 65°C during deprotection.
