Our Customer wants to include a reduced control in the Total Thiol Quantitation Assay (Cat#5524)?

They use TCEP to reduce the protein, but the background signal is high, could you please recommend a way to remove TCEP in solution?

Answer

TCEP is a common reducing reagent widely used to break disulfide bonds within and between proteins. However, there are numerous reports that TCEP is not compatible with downstream assays. Please try ReadiUse™ TCEP removing buffer, which can effectively remove residual TCEP in the reaction mixture and is compatible with the followed maleimide reactions.
