What does primary explant mean?

Answer

Primary explant is a technique developed by Ross Harrison in 1907 for the extraction of cells from tissue. This process involves the following steps:

  • Step 1: Tissue sample is suspended in basal salt solution.
  • Step 2: Carefully slice tissue into pieces and wash by settling.
  • Step 3: Remove the basal salt solution.
  • Step 4: Transfer tissue pieces on to a suitable growth medium.
  • Step 5: Incubate 3 to 5 days and replenish medium as needed.
  • Step 6: Separate explants and transfer to a new culture vessel with fresh growth medium.
