What is the advantage of using the pre-adsorbed secondary antibody?

Posted September 27, 2021

Answer

Pre-adsorption involves removing non-specific antibodies from the mixture. This improves the specificity of adsorbed secondary antibodies, minimizing the risk of non-specific background staining.

Using pre-adsorbed secondary antibodies is highly recommended when working with tissues and cells. They are particularly useful for multicolor experiments where several primary antibodies and their corresponding secondary antibodies are used simultaneously. They are also useful in Immunohistochemistry (IHC) assays on samples with large amounts of endogenous immunoglobulins. In both cases, the pre-adsorption process minimizes the risk of cross reactivity between the secondary antibody and endogenous immunoglobulins present on cell and tissue samples.
