When should ethidium bromide be added to agarose?

Answer

Ethidium Bromide should be added to the running buffer during the separation of DNA fragments by agarose gel electrophoresis. The EtBr molecules become about 20 to 25 times brighter on binding to DNA and fluoresce bright orange when illuminated with UV light, enabling DNA detection.
