Why is DNA fragmented before sequencing?

Posted February 17, 2022

Answer

Sequencing is most successful when performed on fairly short strands of 100 to 5000 base pairs. After a certain length, it becomes difficult to identify the actual base or nucleotide. This is because the quality of the base is inversely proportional to the length of the DNA strand. Subdividing longer DNA sequences into smaller fragments before sequencing provides clearer results.
