Why should I use an absorbance ratio at A575nm/A605nm when using most of your Amplite® Colorimetric Assay Kits?
Posted November 17, 2017

For example catalog# 13838 (Amplite® Colorimetric Glycerol 3-Phosphate (G3P) Assay Kit), catalog# 13803 (Amplite® Colorimetric Alanine Aminotransferase Assay Kit Blue Color), catalog# 13801 (Amplite® Colorimetric Aspartate Aminotransferase Assay Kit Blue Color) etc.

Answer

Compared to performing assay measurements at a single-wavelength, such as 570 nm, we recommend performing a ratiometric technique to increase the sensitivity and accuracy of the assay. Ratioing techniques reduce variables commonly associated with single wavelength indicators such as uneven dye loading, poor dye retention and photobleaching, all of which may obscure your results.
