The binding between the antibody and the target protein may be affected by the coupling of the label to the primary antibody. 
It is less sensitive than the indirect detection technique and may be better suited for applications in which the primary antibody is highly specific and there is a sufficient amount of target proteins. 
Every primary antibody needs to be labeled, which can add to the cost and time.  
It cannot be used for other applications down the line because the primary antibody is tagged with a label.
