What are the limitations of semi-dry transfer techniques for western blot?

Answer

Using semi-dry transfers may not ensure accurate transfer of all proteins for quantitative analysis, particularly those that are extremely small or large. They also may require more effort to optimize sample conditions and re-validate antibodies compared to wet transfers. Additionally, some advanced stain-free detection methods may require specific gel formulations designed for particular instruments. Lastly, certain instruments might have limited capabilities for transfer optimization.
