What are the limitations of solid media for bacterial growth?

Answer

There are various limitations of solid media. One limitation is the shallow nature and extensive surface area of plate media, which can rapidly dehydrate. Thus, it loses its ability to sustain growth within a few days if measures to prevent evaporation are not implemented. Another limitation is that microorganisms exhibit distinct oxygen preferences. In contrast to liquid media, solid media doesn't facilitate the concurrent growth of aerobic and anaerobic bacteria. As a result, duplicate plates are necessary, requiring inoculation and incubation for the two environments. Solid media is limited in accommodating large inocula volumes, typically exceeding 0.05ml.
