During DNA hybridization, the target DNA's hydrogen bonds are denatured either by heating to 95°C or exposure to an alkaline pH, resulting in single-stranded DNA (ssDNA) formation. Subsequently, the ssDNA target is paired with the probe at temperatures around 65°C or lower for an extended period. If the target DNA sequence aligns with the probe sequence, hybridization occurs through hydrogen bond formation.
