Answer

The types of culture media based on physical state include: solid medium, liquid medium, and semi-solid medium.

  1. Solid media is used for isolating bacteria in a pure culture on a solid substrate. Agar (present at a concentration of 1.5-2.0%,) is utilized to solidify the medium. This solid medium facilitates bacterial growth in distinct colonies..
  2. Liquid media supports the abundant growth of bacteria. It facilitates consistent and cloudy growth of bacterial strains when placed in an incubator at 37°C for a duration of 24 hours. Unlike solid media, liquid media lacks agar and is primarily used for fermentation studies.
  3. Semi-solid media is designed for assessing bacterial motility and cultivating microaerophilic bacteria. With agar content typically at 0.5% or lower, it exhibits a gel-like consistency.
