What are the drawbacks of LDH release assay?

Answer

One significant drawback of this assay is that certain compounds contain their own LDH activity, leading to elevated background readings. Consequently, the assay is restricted to serum-free or low-serum conditions, which can shorten the duration of cell culture and the ability to assess cell death under typical growth conditions. Another limitation is the LDH assay exhibits higher intra- and inter-assay variabilities compared to other assays. Additionally, the formulations in these assays contain unknown concentrations of components, which restrict the ability to optimize or modify the assay based on evolving experimental requirements. They also can be expensive.
