β -galactosidase dose response was measured with Amplite® Colorimetric β-Galactosidase Assay Kit in a 96-well clear bottom plate using a SpectraMax microplate reader (Molecular Devices). As low as 3 mU/mL β- galactosidase was detected with 30-60 minutes incubation. Note: The absorbance background increases with time, thus it is important to subtract the absorbance of the blank wells for each data point.

Cell Meter™ Senescence Assay Kits enable direct detection of senescence-associated β-galactosidase (SA-beta-gal) activity in live cells. SA-beta-gal is a well-established biomarker of cellular senescence, an irreversible growth arrest state triggered by DNA damage, telomere shortening, or oncogenic stress. β-galactosidase Substrates

Xite™ Fluorogenic Beta-D-Galactopyranosides
Xite™ beta-D-galactopyranosides are fluorogenic probes designed for β-galactosidase detection with improved fluorescence signal characteristics and cell permeability compared to commonly used substrates. These substrates deliver superior signal-to-background ratios compared to traditional substrates like FDG, with enhanced brightness and improved cell permeability for live cell applications.

FDG, MUP and Other Fluorogenic Substrates
Classic fluorogenic substrates, such as FDG and MUP, provide additional flexibility for researchers developing custom assay protocols or requiring specific spectral properties. These substrates release fluorescent products upon enzymatic cleavage, enabling real-time monitoring of galactosidase activity. Products are available for both alpha- and β-galactosidase detection across ultraviolet, green, and red emission wavelengths.
D-Luciferin Galactoside
D-Luciferin galactoside enables bioluminescent detection of β-galactosidase activity when used in conjunction with firefly luciferase. The beta-galactoside moiety blocks luciferase recognition; upon enzymatic cleavage, free D-luciferin is released and can be quantified through luminescence. This approach is particularly useful for reporter gene assays where luciferase and lacZ are used as dual reporters.
