Human beta-2-microglobulin (B2M) is an 11.6 kDa non-glycosylated protein present in a variety of bodily fluids. It is a component of Class I MHC molecules and is necessary for the proper folding, binding of antigenic peptide, and surface expression of MHC-1 molecules. In long-term hemodialysis patients, B2M can accumulate to cause serious dialysis-related amyloidosis. Additionally, elevated levels of B2M are associated with rheumatoid arthritis, viral infections, cancer and aging. Anti-B2M monoclonal antibodies are highly specific for the detection of human B2M proteins. They can be used in a broad range of immunoassays such as latex enhanced turbidimetric immunoassays (LETIA), lateral-flow immunoassays (LFIA) and ELISA.

Linear calibration curve for B2M in latex-enhanced turbidimetric immunoassay (LETIA): human B2M was reacted with the anti-human B2M antibody-coated latex, resulting in agglutination and an increase in turbidity. Turbidity changes were monitored using a spectrometer to quantitatively measure the B2M concentration in the sample.

Clinical comparison of AAT Bioquest’s B2M reagent and commercial LETIA kit: a B2M lateral-flow immunoassay (LFIA) using anti-B2M antibody (Cat# V100015; labeled with gold) and capture antibody was compared with a commercial B2M LETIA kit. Each assay was performed using 6 blood samples which were diluted by 50-fold.
