Interleukin 6 (IL-6) is a pleotropic cytokine encoded by the IL6 gene in humans. It acts as both a pro-inflammatory cytokine and as an anti-inflammatory myokine. IL-6 is essential for stimulating acute phase protein synthesis, hybridoma growth, bone metabolism, inflammation and cancer progression. In almost all diseases involving chronic inflammation, such as diabetes rheumatoid arthritis and congestive heart failure, the concentration of IL-6 is elevated. Anti-IL-6 monoclonal antibodies are highly specific for the detection of human IL-6. They can be used for a broad range of immunoassays such as chemiluminescent immunoassays (CLIA) and rapid sandwich immunoassays.

Figure 1. Comparison of IL-6 CLIA and commercial diagnostic assays: 23 samples from healthy donors and patients with chronic infections were detected using MAb combination B (Cat# V100135 (Clone 2) - Cat# V100135 (Clone 4)) in CLIA IL-6 assay and a commercial diagnostic assay. The correlation coefficient between CLIA IL-6 assay and comparison assay is over 0.97. Results show that these monoclonal antibodies can be applied to double-MAb-sandwich-immunoassays.
Figure 2. Six-point calibration curve for IL-6 sandwich chemiluminescence immunoassay (Calibrator concentrations: 0, 2.5, 25, 250, 750, 1500 pg/mL). All MAbs were tested in pairs as capture and detection antibodies to select the best two-site MAb combinations for the development of a quantitative sandwich immunoassay. Detection antibodies were labeled with horseradish peroxidase (HRP). The best selected MAb combinations for the development of quantitative human IL-6 immunoassays are (capture-detection respectively): MAb combination A: Cat# V100135 (Clone 1) - Cat# V100135 (Clone 4); MAb combination B: Cat# V100135 (Clone 2) - Cat# V100135 (Clone 4); MAb combination C: Cat# V100135 (Clone 3) - Cat# V100135 (Clone 4); MAb combination D: Cat# V100135 (Clone 4) - Cat# V100135 (Clone 2).
