Prolactin (PRL), also known as luteotropic hormone, is a protein secreted from the pituitary gland. It plays a vital role in metabolism, immune system regulation and pancreatic development, but is best known for its role in producing milk in mammals. In normal individuals, PRL concentrations increase in response to physiologic stimuli such as eating, mating, exercise, stress and hypoglycemia. PRL levels are also elevated during pregnancy, lactation, postpartum and in newborn infants. In clinical biology, prolactin tests are used in the diagnosis of prolactinoma, a prolactin-producing tumor of the pituitary gland. Anti-PRL monoclonal antibodies are highly sensitive for the detection of human purified prolactin. They can be used for a broad range of immunoassays such as chemiluminescent immunoassays (CLIA) and in the development of highly sensitive sandwich immunoassays.

Figure 1. Clinical comparison of AAT Bioquest’s prolactin immunoassays and the Access Prolactin assay from Beckman Coulter: 80 clinical blood samples were separately tested using MAb combination A and B on AAT Bioquest’s CLIA platform and compared to a diagnostic kit from Beckman Coulter. Data from this study were analyzed using the Passing-Bablok regression method and are summarized in the following scatter plot. Results show good agreement between AAT Bioquest’s immunoassays and comparison assays.

Figure 2. Calibration curves for prolactin in sandwich chemiluminescence immunoassay (CLIA). All monoclonal antibodies were tested in pairs as capture and detection antibodies to determine the best two-site MAb combinations for the development of a quantitative sandwich immunoassay. Detection antibodies were labeled with horseradish peroxidase (HRP). The best selected MAb combinations for quantification of human prolactin are (capture-detection): MAb combination A: Cat# V100160 (Clone 1) - Cat# V100160 (Clone 2)ï¼›MAb combination B: Cat# V100160 (Clone 1) - Cat# V100160 (Clone 3).
