Serum amyloid A1 (SAA1) is a major acute-phase protein. It is produced primarily by hepatocytes in response to proinflammatory cytokines such as tumor necrosis factor α (TNF-α) or interleukin 6 (IL-6). Under inflammatory conditions, such as infection or malignancy, plasma concentrations of SAA1 can increase as high as 1000-fold. As such, it has become a promising reliable clinical indicator and biomarker for inflammatory diseases, chronic disorder and late-stage malignancy. Anti-SAA1 monoclonal antibodies are highly sensitive for the detection of human serum amyloid A1 immunogens. They can be used in the development of a broad range of immunoassays including gold immunochromatographic assays (GICA) and latex enhanced turbidimetric immunoassays (LETIA).

Figure 1. Clinical comparison of AAT Bioquest’s LETIA immunoassay and a competitor’s immunoassay: anti-SAA1 monoclonal antibodies were evaluated in medium-scale clinical tries with 100 blood samples from donors. The correlation coefficient (r) is as high as 0.93 between in-house latex reagents and commercial SAA1 immunoassay showing good agreement between the two systems.

Semi-quantitative detection of SAA1 protein in colloidal gold immunochromatogragphic assay (GICA): a set of SAA1 calibrators with the concentration of 0, 0.5, 2, 10, 20, 50, 100 and 200 mg/L was detected on a LFIA platform using two anti-SAA1 monoclonal antibodies from AAT Bioquest. The capture antibodies were stripped on the nitrocellulose membrane, and the detection antibodies were conjugated to colloidal gold. The best selected MAb combination for the development of semi-quantitative human SAA1immunoassays is (capture-detection): Cat# V100140 (Clone 2) - Cat# V100140 (Clone 5).

Calibration curve of SAA1 in particle-enhanced turbidimetric immunoassay: SAA1 antigens were reacted with anti-human SAA1 monoclonal antibodies pre-coated on latex beads, resulting in agglutination and increase in turbidity. Changes in absorbance were detected using a biochemical analyzer.
