Cyclin-dependent kinase inhibitor 1 (sometimes referred to as MDA-6, Melanoma differentiation-associated protein 6, p21 or CDK-interacting protein 1) is a protein with a molecular weight of 18 kDa, found in the PCNA-p21 complex, nucleoplasm and cytosol of cells. In humans, CDK-interacting protein 1 has been found to be involved in cyclin-dependent protein serine/threonine kinase activity, cell cycle G1/S phase transition and transcription by RNA polymerase II. It is the subject of extensive research stemming from the fact that it is a member of the intrinsic apoptotic signaling pathway, cytokine-mediated signaling pathway and intrinsic apoptotic signaling pathway in response to DNA damage by p53 class mediator. It negatively regulates cardiac muscle tissue regeneration, phosphorylation and cell population proliferation. On the other hand, it also enhances programmed cell death, B cell proliferation and fibroblast proliferation. CDK-interacting protein 1 binds to protein-containing complex, protein kinase and metal ion. It plays an important role in organismal processes, namely, stress-induced premature senescence, mitotic G2 DNA damage checkpoint and DNA damage response, signal transduction by p53 class mediator resulting in transcription of p21 class mediator.
