Protein piccolo is a 561 kDa protein that can be expressed in the extracellular exosome, cytoskeleton and GABA-ergic synapse of cells. In Homo sapiens, protein piccolo aids in organismal processes, for example, cAMP-mediated signaling, synaptic vesicle clustering and cytoskeleton organization. Sequencing of protein piccolo has supported it contains 3 types of conserved structural units: PDZ, C2 1 and C2 2 domain. It is the subject of comprehensive study in part because of the fact that it is a component of the presynapse to nucleus signaling pathway. It is an integral part of exocytosis. Protein piccolo binds with profilin, calcium-dependent phospholipid and calcium ion. It is clinically significant because abnormalities in its function have been thought to be involved with diseases like pontocerebellar hypoplasia type 3 (PCH3). Pontocerebellar hypoplasia type 3, an autosomal recessive inheritancedisorder characterized by cerebral atrophy, macrotia and proptosis, has in particular been of interest to investigators.
