Suppressor of cytokine signaling 3 (sometimes called SSI-3, Cytokine-inducible SH2 protein 3, STAT-induced STAT inhibitor 3 or CIS-3) is a protein with a molecular weight of 25 kDa, located in the phosphatidylinositol 3-kinase complex and cytosol of cells. In humans, suppressor of cytokine signaling 3 has been associated with key functions such as protein kinase inhibitor and 1-phosphatidylinositol-3-kinase regulator activity. It plays a role in the upregulation of tyrosine phosphorylation of STAT protein and cell differentiation but also is an inhibitor of tyrosine phosphorylation of STAT protein, apoptotic process and inflammatory response. Sequencing of suppressor of cytokine signaling 3 has shown it contains 2 types of conserved structural units: SOCS box and SH2 domain. Suppressor of cytokine signaling 3 binds to phosphotyrosine residue. Suppressor of cytokine signaling 3 is the subject of comprehensive research because it plays a role in the receptor signaling pathway via JAK-STAT, cytokine-mediated signaling pathway and regulation of interferon-γ-mediated signaling pathway. It is an integral part of organismal processes, for example, protein ubiquitination, cellular response to leukemia inhibitory factor and post-translational protein modification.
