Non-structural polyprotein pORF1 is a protein with a molecular weight of 216 kda, found in the host cell cytoplasm of cells. In hev-1s, non-structural polyprotein pORF1 has been associated with essential functions like helicase, mRNA methyltransferase and RNA-directed 5'-3' RNA polymerase activity. Sequencing of non-structural polyprotein pORF1 has shown it contains 5 conserved structural units: alphavirus-like MT, RdRp catalytic, Macro, (+)RNA virus helicase C-terminal and (+)RNA virus helicase ATP-binding domain. Non-structural polyprotein pORF1 binds with identical protein, RNA and ATP. It has been found to be involved in organismal processes, namely, viral protein processing, DNA-templated transcription, and viral RNA genome replication.
