How do you determine the concentration of peridinin-chlorophyll protein complex (PerCP)

Answer

To determine the concentration of PerCP you will need to know these values:

  • the molecular weight of PerCP = ~35.5 kDa
  • the λex of PerCP = 482 nm in PBS
  • the extinction coefficient of PerCP = ~400,000 cm-1M-1
  • the extinction coefficient E1% (482nm) of PerCP = 116
    • (absorbance of 10 mg/ml solution in a cuvette with a 1 cm path length is 116)

 To calculate PerCP concentration use the following:

 PerCP concentration (mg/ml) = (Abs482 nm  measurement) / 11.6
