The probe provided in your GSH/GSSG ratio assay (Cat#10060) only reacts with reduced GSH, not GSSG. Why do I also need to prepare a GSSG standard curve?

Answer

During oxidative stress, GSSG accumulates in cells increasing the ratio of GSSG to GSH. Since Thiolite™ Green 520WS only reacts with GSH, the GSSG that accumulated must first be enzymatically converted to GSH. Therefore, to measure GSH and Total GSH you must create a GSH and a GSSG standard curve. Reduced GSH is measured using the GSH standard and Total GSH (GSH and GSSG) is measured using a GSSG standard. To calculate GSSG, you subtract the GSH amount from the Total GSH, and then you use this value to determine your final GSH/GSSG ratio.
