RNA-sequencing involves converting an RNA sample to a cDNA library, which is then sequenced and mapped against a reference genome.

The process of RNA-sequencing consists of the following steps:

  • Designing experiment – The experiment is set up to address the researcher’s specific questions.
  • Preparing the RNA – The input RNA is isolated and purified.
  • Preparing the Libraries – The purified RNA is converted to cDNA and sequencing adapters are added.
  • Sequencing – cDNAs are sequenced using a sequencing platform.
  • Analysis – The resultant short-read sequences are analyzed.
