How do I dilute protein for BCA assay?

Answer
  1. Determine the Required Concentration: Based on the expected protein concentration and the linear range of the BCA assay, decide on the final concentration of the protein that will be used in the assay. Typically, this falls within the range of 0.5 to 10 mg/mL for the BCA assay.
  2. Prepare the Diluent: Use the same buffer or solvent in which the protein sample is dissolved to avoid introducing contaminants. This could be the buffer used in the protein’s storage.
  3. Calculate Dilution Factor: To achieve the desired concentration, calculate the dilution factor needed. 
    1. Dilution Factor= Initial Concentration / Final Concentration 
  4. Prepare the Diluted Sample: Mix the protein sample with the diluent according to the dilution factor.
