What are the advantages of reduced representation bisulfite sequencing (RRBS)?

Posted April 26, 2024

Answer

One advantage is that RRBS enables cost-effective sequencing of CpG-rich regions across the genome at single-base resolution compared to WGBS. This is since RRBS concentrates on CpG-rich regions of the genome. Accurate data analysis is achievable even with low sample concentrations. RRBS can identify both inter- and intragenic differentially methylated regions (DMRs) with high resolution and can detect methylation changes in coding and non-coding genome regions. Additionally, it's highly sensitive, needing only 10 ng or more of non-degraded, high-quality genomic DNA. By targeting CG sites, RRBS can capture methylation patterns in regions that are likely to be functionally important, such as promoter and enhancer regions. RRBS also allows for the detection of tissue-specific methylation patterns by isolating and analyzing specific cell types.
