What are the disadvantages of Chemiluminescent detection method for western blot?

Answer

One disadvantage is that Chemiluminescent detection cannot simultaneously detect two similar-sized proteins (multiplexing). Sequential detection is necessary, which involves stripping the membrane of the first detection antibody before incubating it with the antibody against the second target protein. Membrane stripping necessitates extra time and functions to prevent protein loss. Another limitation is that light emission occurs only when the substrate converts into a product, requiring rapid detection following substrate addition.
