What are the disadvantages of colorimetric detection for western blot?

Answer

One disadvantage is they are challenging to quantify accurately since the individual determines the precipitation reaction's time span, and is often irreversible. The assay also has a restricted linear range. Another disadvantage is that it is less sensitive than fluorescent and chemiluminescent detection methods, typically requiring the presence of nanograms of protein for visible bands.
