Are there any disadvantages of using mounting medium in biology?

Answer

The major disadvantage of using mounting medium in biology is that samples must be dehydrated before mounting. This can be hugely time-consuming with some histological moutants requiring excessive dehydration steps to prepare the sample for mounting. Another disadvantage is that over time, the clearing action of the medium may produce a specimen with the same refractive index of the mounting media, rendering the specimen undetectable under the microscope.
