What are the advantages of single molecule real time sequencing?

There are several advantages of single molecule real time sequencing (SMRT). One advantage is that it minimizes chemical modifications during library preparation, eliminating the need for DNA amplification. Another pro is that it allows for the detection of various types of epigenetic modifications. Moreover, it produces longer reads, averaging around 3000 base pairs. This feature enhances the assembly of entire genomes, as longer reads reduce the complexity of the assembly process. This also increases the chances of detecting sequences present in the sample at lower abundance. SMRT also relies on a DNA polymerase to facilitate the reaction and utilizes single-molecule imaging. Because of this, there is no gradual decline in signal strength over time and instead the sequencing process concludes when the polymerase and template separate.
