What are the disadvantages of SMRT sequencing?

Answer

There are several disadvantages of SMRT sequencing. The main disadvantage is the low-throughput. For example, PABICO has a lower throughput of the flow cell compared to the Illumina platform. The issue arises from the fact that not all Zero-Mode Waveguides (ZMWs) on the flow cell carry out successful sequencing reactions. Some wells lack the DNA template. Another challenge is the relatively high sequencing error rate associated with PACBIO compared to the Illumina platform. Currently, achieving 99% accurate data necessitates a minimum of 15 passes. This significantly impacts both the cost and time involved in the sequencing process.
